The concentration of insulin in serum-free medium is generally between 5–20 mg/L. The most suitable concentration should be selected for different cell lines.
Insulin is supplied as a crystalline powder and should be stored frozen below -18°C. Before use, dissolve it, typically preparing a stock solution of 1–2 mg insulin per 1 ml using hydrochloric acid solution (10 mM), then sterilize by filtration. The filtered solution can be stored at -20°C for 6 months after aliquoting. Avoid repeated freeze-thaw cycles.
